Three options:

- By train leaves from Santiago at 13:45 and arrives to Madrid at 22:00.
Another one goes by night, leaving at 22:30 and arriving to Chamartin
Station in Madrid at 07:30 (www.renfe.es)
- By bus (www.alsa.es).
- By plane, much expensive but quickler too.
